I have just purchased a 2010 premium luxury 3l diesel & am unsure when Sport mode should be selected. To me, the handbook is not clear on this point. Can you select Sport instead of Drive after starting the engine or should you switch from Drive to Sport when the car is in motion?

Hi and welcome to the forum. Sports mode is a personal preference. I assume it is selected after starting the engine and resets to normal when turning the engine off.

You can select Sport mode either way, before you set off or while in motion. But obviously not when in Reverse!
Once Sport is selected you must use the right paddle to change up gears otherwise it will hold the selected gear and rev to the limiter, but it down changes by itself as you slow right down to a stop.
Use Sport if you want a «sportier» driving experience using the paddles and select Dynamic as well if you want a bit more verve.
You can also simply use the paddles while in D, it doesn’t need to be in Sport to use them, but in D after a while of no paddle action it will change up to a more «relaxed» gear by itself.

You don’t need to use the paddles in Sport unless you want to, as it changes up & down itself — it just hangs on to gears longer so you’re often in a lower gear than when using Drive. Top gear is also unavailable in Sport.
Now, I’ve heard that different XF models do different things, depending on model & factory software. Mine is a late 2009 XF 3.0 litre Diesel S. Even if I use the paddles in Sport mode, if I make no changes for roughly 40 secs, it reverts to auto. Only when I’m in Sport, with Dynamic mode on & I then use the paddles will it stay in manual mode until I cancel it.

As Robinus — they all seem to differ slightly. On my 2012 S, in sport if I use the paddles it locks into manual until I cancel it (hold the paddle). Outside that, it just holds lower gears. No need for Dynamic. That just adds stiffer suspension and a sharper throttle.
Its all software at the end of the day so you can swap around between settings when and how you want with impunity. If it’s not mechanically safe, the car will just refuse.

I use the sport mode for overtaking as the box is more responsive to change down a cog or two particularly in paddle mode. Not that I use the paddles much — I think the XF may have made me a lazier driver content to let the auto box do the work most of the time.

I usually drive with a light foot for overall fuel economy however find that when sharper response is required the car can appear a bit sluggish in its response — due I think to the learning capability of the gear box which has adapted to gentle treatment. Does use of Sport mode short circuit the learning process to give more immediate response or is it that it just drops down a gear or two? When I first had the car I thought I would use the paddles a lot but found that the car was so good at changing just when I would that I too became lazy relying on the auto. Sport is good for a bit of fun but driving with the paddles in a more spirited way seems to be just as effective as the gearbox seems to adapt quite quickly. What exactly does sport mode do?
